บทคัดย่อ/คำอธิบาย :

A three dimensional-quantitative structure activity relationship (3D-QSAR) model has been developed to rationalize the effectiveness of mitragynine (MG) analog that will be synthesized. 7-hydroxy mitragynine (7-0HMG) has been selected as the analog to be studied further, thus a synthetic reaction to produce 7-0HMG from MG has been developed and optimized with an almost 100 percent conversion. The analgesic results also proved that 7-0HMG is more potent than morphine that may due to its higher lipophilicity and easy penetration to the blood brain barrier. However when 7- OHMG is converted to its CDS of nicotinic ester that contains nicotinic acid group which is believed to penetrate blood brain barrier better through a nicotinic transport system in the brain, show lower activity when compared to 7-0HMG may due to its lower solubility in water. Sodium and potassium salts of 7-0HMG have also been successfully synthesized and showed greater solubility in water than the 7-0HMG.
